چکیده

Dopamine-fS-hydroxylase (DBH), the enzyme responsible for the conversion of dopamine to norepinephrine, was measured in the plasma of two lines of mice selectively bred for differential sensitivity to hypnotic doses of ethanol. Plasma levels of DBH were measured by radioenzymatic assay in long sleep (LS) and short sleep (SS) male and female mice which had no prior exposure to ethanol. LS mice had ,significantly higher plasma DBH activity than did SS mice, although no significant sex difference was found. Fusaric acid, an inhibitor of both central and peripheral DBH, attenuated sleep time induced by either 3 or 4 g/kg ethanol in mice of a heterogeneous genetic stock. The results of these experiments may further implicate noradrenergic systems in mediating some of the depressed effects of ethanol.

10. Pharmacology of Isoquinoline Alkaloids and Ethanol Interactions*

Very few of the more than sixty naturally-occurring isoquinoline alkaloids have been screened for pharmacological activity, although certain members of this class have known hallucinogenic and cardiovascular activity. Recently the suggestion that biosynthetic catecholamine-derived tetrahydroisoquinolines are involved in the etiology of alcoholism has stimulated research into their activity. Bot...

Implications of endogenous opioids and dopamine in alcoholism: human and basic science studies.

We investigated the endogenous opioid system and its role in mediating the reinforcing effects of ethanol that lead to high ethanol consumption as a biochemical marker of an individual's vulnerability to excessive ethanol consumption. We performed studies using human subjects with [high risk (HR)] and without [low risk (LR)] a family history of alcoholism to supplement our studies with experime...

Mice selectively bred for high- or low-alcohol-induced locomotion exhibit differences in dopamine neuron function.

Elevated sensitivity to the euphoric or stimulant effects of ethanol is associated with higher levels of alcohol use in some human populations. Midbrain dopamine neurons are thought to be important mediators of both ethanol reward and locomotor stimulation. Patch-clamp recordings were used to examine the electrical properties of dopamine neurons in a genetic model of heightened (FAST) and reduc...

Anaesthetic requirement in mice selectively bred for differences in ethanol sensitivity.

Anaesthetic requirements for nitrous oxide, enflurane and isoflurane were determined in mice selectively bred for their susceptibility ("long-sleep" mice) or resistance ("short-sleep" mice) to alcohol. Nitrous oxide and enflurane requirements, measured by the rolling-response test, were 34 and 20% greater, respectively, in short-sleep mice than in long-sleep mice. Although isoflurane requiremen...

Ethanol- and cocaine-induced locomotion are genetically related to increases in accumbal dopamine.

Neuroanatomical research suggests that interactions between dopamine and glutamate within the mesolimbic dopamine system are involved in both drug-induced locomotor stimulation and addiction. Therefore, genetically determined differences in the locomotor responses to ethanol and cocaine may be related to differences in the effects of these drugs on this system. To test this, we measured drug-in...
